The traditional Yeast Two-Hybrid system works in the nucleus and is unsuitable for membrane proteins. This system uses split-ubiquitin or other technologies to reconstitute transcription factors upon interaction at the membrane, allowing for the discovery of protein-protein interactions for full-length membrane proteins in their native cellular context.
