Discover how the Thermo Scientific ToxInsight DILI Assay Cartridge can prevent you from finding undesirable liver toxicity in your lead compounds late in the discovery process. The ToxInsight DILI Assay Cartridge identifies drug induced hepatotoxicity by simultaneous detection of multiplexed targets including cell loss, nuclear DNA, reduced glutathione level, intracellular reactive oxygen species and mitochondrial membrane potential.

The number of new drugs approved for use has been in sharp decline for the last decade, despite large investments in pharmaceutical R&D. While there are a variety of reasons behind this attrition rate, lack of efficacy and undesirable toxicity are two major reasons for this productivity gap.

Drug Induced Liver Injury (DILI) is the number one reason for post market drug withdrawal and now a cell based, predictive DILI detection assay is available as a cartridge on the Thermo Scientific ToxInsight IVT platform. The ToxInsight DILI Assay Cartridge identifies drug induced hepatotoxicity by simultaneous detection of multiplexed targets including cell loss, nuclear DNA, reduced glutathione level, intracellular reactive oxygen species and mitochondrial membrane potential. The ToxInsight DILI Assay Cartridge includes all the reagents, assay protocol, analysis and data reporting capabilities to predictively profile compounds for hepatotoxicity with high sensitivity and specificity. The cartridge has been extensively validated on a range of hepatic cell-lines including rat and human primary hepatocytes, with a known hepatotoxic compound “truth-set”.

Key to the predictivity of the DILI Assay Cartridge is the data analysis template that uses powerful statistical algorithms and visualization to make sense of the multi-parameter data from the cartridge. The template supports analysis and toxicity prediction of compounds with broad mechanisms of action because it monitors multiple, independent toxicity indicators. Intuitive visualization tools are built into the template that provides the user with graphs of individual responses for selected compounds and a Boolean table of predicted toxicity so the user can swiftly determine a compound’s risk of liver injury in minutes.

The Thermo Scientific ToxInsight IVT platform offers a predictive, cell based imaging approach to determining the toxicity risk of a compound through the measurement of multiple toxicity biomarkers in individual cells. The automated, bench-top, easy to use platform allows a researcher to benefit from physiologically relevant toxicity assays early in the drug discovery process, rather than traditional late stage histopathology or animal studies.

The ToxInsight IVT platform is designed to both profile compounds for their toxicity risk and then determine the mechanism of action of any toxicity detected, enabling researchers to rapidly understand the therapeutic index of a compound series in minutes. Compared to traditional biochemical assays (such as cell count, LDH, MTT etc) the ToxInsight IVT platform makes multiple measurements on each cell generating a phenotype rather than a single measurement.
