Thermo Fisher Scientific Introduces Alternative to Phospho-Antibodies for the Study of Phosphotyrosine Signaling

Thermo Fisher Scientific, the world leader in serving science, now offers the Thermo Scientific SH2 Domain Phosphotyrosine Capture Kits, fast and efficient tools for capturing specific tyrosine phosphorylated proteins from mammalian cell lysates. These new kits enable researchers to monitor and profile specific tyrosine phosphorylation events and identify specific SH2 domain protein-protein interaction networks.

The non-antibody-based assay uses GST-tagged SH2 domain peptides and glutathione agarose resin to capture and pull down the specific binding partners, providing an alternative to traditional immunoprecipitations using phospho-antibodies. The SH2 Domain Phosphotyrosine Capture Kits also offer an alternative to phosphoprotein enrichment resins that do not discriminate among proteins that are phosphorylated on serines, threonines or tyrosines. Subsequent analysis of the SH2 domain interactions can be performed using Western blots or mass spectrometry.

Kits for studying 17 specific SH2 Domains are available. Each kit contains 600 µg of a specific GST-SH2 domain and sufficient reagents (including negative controls) for six pull-down reactions with approximately 500 µg of lysate. The kits have a shelf life of one year.
