Three isotype-specific antibodies are detected: IgA, IgG, and IgM, providing the spectrum of antibody response over time after COVID-19 vaccine administration or natural exposure to SARS-CoV-2.

Human samples require four components for a complete assay:
- Antigen-coupled beads
- Isotype-specific detection antibody
- Isotype-specific control set

For samples from other species, researchers must provide their own biotinylated anti-species detection antibody, positive and negative controls, and select a reagent kit appropriate for the chosen species.
