Organ-on-Chip Modeling of Cancer Therapeutic Response in a Vascularized and Immune-Competent Microenvironment

I have used S1 Chips to develop a prostate cancer-on-a-chip model to study tumor response to therapy in the context of immune interactions under the control of vascular dynamics. Using the dual-channel design, I established a compartment of vascular endothelial cells to mimic the in vivo vasculature. I introduced immune cells in the adjacent channel to simulate their trafficking and interaction with tumor cells. This setup enabled me to observe treatment-induced changes under more physiologically relevant conditions. The platform provided the flexibility to adapt complex co-culture systems and the robustness to support long-term studies. With the support of Emulate’s technical team and detailed documentation, I was able to streamline protocol optimization and focus on capturing meaningful biological data. Looking ahead, I am interested in further improving the experimental design to study the immune system’s role in greater detail. This includes incorporating downstream flow cytometry to profile immune cell lineages and functional changes, and proteomic analysis of secreted factors to understand cellular communication across compartments better. Overall, the S1 Chips have significantly advanced my ability to model therapeutic responses in a microenvironment that can closely mirror human prostate cancer biology.

Lung-on-a-Chip Modeling of Ischemia Reperfusion Injury
I've used this system for 3 years performing lung-on-a-chip experiments with the S1 chips. We began with the validated protocol provided by Emulate and, with the help of their professionals, added elements to create a new and more representative cell culture model of lung ischemia reperfusion injury. Using the CM2 culture module allowed us to jump over the initial hurdles of designing and troubleshooting an in-house organoid system so that we could quickly begin addressing the more interesting scientific questions relevant to our research. Additionally, by using a customer facing system we hope that this will improve the replicability and impact of our work.

The Chip-S1 consumable enables users to build robust organ models with a tissue-vascular interface and has been validated for a variety of organ models and applications. Kit components include Chip-S1 Stretchable Chips, Pod® Portable Modules, ER-1® / ER-2®, and Steriflip Filters. Users will need to supply their own biological components (cells, ECM, and media). Organ-Chips created using this kit can be cultured on the Human Emulation System for precise control of culturing conditions (media flow rates and stretch frequency).
