Specifically designed for use in ACEA’s xCELLigence® RTCA DP instrument, our Cell Invasion and Migration plate (CIM-Plate®) is a 16-well electronically integrated transwell device composed of upper and lower chambers that easily snap together.
A polyethylene terepthalate (PET) microporous membrane serves as the base of the upper chamber, allowing for cells to translocate towards chemoattractant in the lower chamber. The bottom side of this membrane is coated with gold microelectrode sensors that have a reduced ability to conduct electric current when cells adhere to their surface. It is this real-time “impedance” signal which enables a quantitative kinetic measurement of cell movement from the upper chamber to the lower chamber.
While for a standard migration assay cells are seeded directly on top of the membrane in the upper chamber, for invasion assays cells are seeded atop an extracellular basement membrane matrix (such as Matrigel) or a cellular monolayer previously established on the membrane. The use of impedance to measure cell invasion/migration enables a high level of efficiency, reproducibility, and throughput that is simply not possible using traditional methods such as a Boyden chamber. Once an experiment is completed, impedance data can be corroborated simply by staining the CIM-Plate membrane and visualizing translocated cells under a microscope.
