BACKGROUND
Following an initial discussion and comprehensive review of the literature, our molecular biologists will use a range of bioinformatics tools to design expression constructs. Different strategies and approaches will be considered, including (but not limited to) expression host, purification tags, signal peptide sequences (secreted targets), domain boundaries, mutagenesis and end-use of the protein. Once an optimal construct has been found, recombinant DNA will be produced by gene synthesis and cloned into a suitable vector, using established molecular biology techniques. Constructs will be sequence-verified upon completion. Expression can be carried out in parallel in different hosts, including E. coli, baculovirus-insect cells and mammalian cells to obtain the highest yields and most authentic post-translational modifications. Initially, small-scale expression trials will be carried out to confirm optimal conditions, before scaling up to large-scale production. We utilize a broad range of purification methods to achieve maximal protein purity and consistency. Purifying your protein of interest will begin with a discussion about your specific requirements, which may include functional protein purity, enzymatic activity, removal of specific contaminants or the use of bespoke buffers. Following consultation, we work with you to develop a custom strategy for purification. An initial pilot-scale purification may be recommended in some instances to optimize the process prior to large-scale purification. Our column chromatography suite includes state-of-the-art AKTA equipment and a full range of chromatography techniques. Following purification, we can also offer to carry out extended QC, including mass spectroscopy, circular dichroism, confocal microscopy, electron microscopy, analytical ultracentrifugation, N-terminal sequencing and endotoxin testing.
The antigens we produce can be used to develop high-quality and target-specific polyclonal, monoclonal or engineered antibodies. We strive to not only provide best-in-class services, but offer custom solutions that can meet your bespoke requirements. We initially arrange a consultation with one of our antibody experts and advise on the process that offers you the best chance of raising an antibody that performs to your desired specifications. After successful completion of a project we can offer a range of additional services, including storage of bulk antibody, hybridoma banking, bulk antibody production, conjugation of the antibody to enzymes or fluorochromes and a range of antibody engineering options including chimerization.
We have many years of experience in assay development. Depending on the application, we advise and test multiple different ELISA formats from simple direct ELISAs to indirect ELISAs with blocking antigens and double antigen-binding ELISAs. Not only do you get the benefit of consultancy with our senior assay development scientists, but our in-house expression systems can also be used to produce natively-folded and fully glycosylated proteins and antibodies to serve as highest quality unique reagents, giving your ELISA the edge against the competition.
