Multicellular 3D spheroids followed by FFPE for IHC based study
1. Easy to use. 2. I could grow my 3D spheroids for 25 days and expect it to maintain the spheroids for 15 more days. 3. My downstream application was to prepare FFPE followed by IHC study. For that, I succeeded in preparing FFPE blocks, H&E and doing IHC of some target proteins. I could see uniform-sized spheroids using these plates. The integrity of the spheroids remains mostly intact after so many stages (e.g. parformaldehyde fixing, low melting agarose embedding, FFPE block preparation, sectioning. For H&E its a few steps but for IHC there are multiple steps of washing and most of the spheroid's structure i.e. round shape still remained intact, so that is a plus). 4. My only concern is value for money. I have used other commercially available plates, although none of them can produce equal-sized spheroids, but to be honest they are not bad. I can conclude any advantage or disadvantage of equal-sized spheroids once I do the hypoxic marker(s) staining.
