A rapid and sensitive homogeneous assay for cortisol assessment

The HTRF® cortisol assay can be used for the quantification of cortisol in rodent serum, or directly on cells. This assay has also been validated for 11-beta HSD1 screening on whole cells or microsomes.

The assay requires only 2 reagent additions, followed by an incubation period of 2 hours at room temperature. It has a detection limit of 70 pg/mL (e.g. 2 fmol/well in 20 µL final volume) for sample cortisol concentrations ranging from 0.1 to 100 ng/mL. The low cortisone cross-reactivity (0.9%) allows the assay to be run in the presence of cortisone, the substrate of 11b-HSD1. Miniaturization to a 384- or 1536-well format does not affect the assay quality, and the test is particularly well suited to HTS with Z’ factors reaching 0.9.
