We have used the Janelia Fluor 646 HaloTag Ligand in our lab successfully. It is easy to use and a great reagent for live imaging. We find that, contrary to the protocol, aliquoting and freezing works fine.

Brighter Ligands, No-Wash Protocol
The Janelia Fluor® 549 HaloTag® Ligand and Janelia Fluor® 646 HaloTag® Ligand enable characterization of HaloTag® fusions in endogenous cellular settings. The enhanced brightness of the dyes associated with the HaloTag® ligands enables their use in detection and single-molecule imaging studies in live cells via:
- FACS
- Standard confocal imaging
- In-gel detection with a fluoroimager
