I've been trying different transfection reagents; however, in my hands jetPEI is one of the best one. It's very simple to use. First of all you need to try to optimize the N/P ratio or jetPEI/DNA complexes. The complexes are added to the cells in serum-containing medium. Depends on your experiment, the reporter gene assay can be performed after 24 to 72 hours after transfection.
