The Lumit™ FcRn Binding Immunoassay is a competition assay to measure the interaction between human FcRn and Fc proteins, including antibodies. A Human IgG1 labeled with LgBiT (Tracer-LgBiT) is used as the tracer. A C-terminal biotinylated human FcRn bound to Streptavidin-SmBiT is used as the target. In the absence of an antibody analyte sample, Tracer-LgBiT binds to the hFcRn-SmBiT target resulting in a maximum luminescence signal. Unlabeled IgG will compete with Tracer-LgBiT for binding to the FcRn target, resulting in a concentration-dependent decrease in luminescent signal. It's very efficient and effective. Also, very fast to use. In comparison to other technologies likes ELISAs, Lumit™ FcRn Binding Immunoassay allows users to maximize data obtained from a minimum volume of sample. For many ELISAs, it requires 100 microliters or more to get one answer for one specific analyte, but with multiplexing, depending on how large a panel is, you are able to get a lot of information from very minimal samples (somewhere around 10–13 microliters, most of the time). A lot of researchers who work with animal models like mice—from which you get very low sample volume to begin with—really appreciate the fact that you can get so much data from such a small sample volume. Lumit™ FcRn Binding Immunoassay also increases throughput by allowing you to save some time. Many available immunoassays will require an incubation of anywhere between a couple of hours to overnight. For Bio-Plex assays, the primary incubation is between 30 minutes and an hour, and, overall, you can get data in as little as three to four hours. That is a big advantage for people who want data quickly. Also, the fact that you can get all these answers for many different analytes at the same time allows for more consistent data. And because you do not add confounding variables like different operators or different buffer systems, you really get a snapshot of the profile you are looking at, whether it be cytokines or other proteins.

In analyte samples, unlabeled IgG will compete with Tracer-LgBiT for binding to the FcRn target, resulting in a concentration-dependent decrease in luminescent signal.
