Prostaglandin E2 (PGE2) is a primary product of arachidonic metabolism in many cells. Under inflammatory stimuli, PGE2 is de novo synthesized by three enzymatic steps via the cyclooxygenase (COX) pathways. PGE2 production is commonly used in the detection of COX-1 and COX-2 activity.
In the PGE2 HTRF® assay, free PGE2 from the sample competes with XL665 labeled PGE2 for binding to a Cryptate conjugated anti-PGE2 antibody. This competitive homogeneous immunoassay only requires the mixing of the stimulated-cell supernatant with the two conjugates, incubation for 4 hours at room temperature and then fluorescence readout. Detection limit is below 20 pg/mL, with a signal to background ranging from 6 to 10, depending on incubation conditions.
