• A DNA sequence encoding the mature human IL-6 protein sequence (Hirano, T. et al., 1986, Nature 324:73 - 76) was expressed in E. coli.
Molecular Mass of R&D IL-6
• The 184 amino acid residue recombinant protein has a predicted molecular mass of approximately 20.3 kDa.
Purity of R&D IL-6
• > 97%, as determined by SDS-PAGE and visualized by silver stain.
Endotoxin Level
• < 1.0 EU per 1 μg of the cytokine as determined by the LAL method.
Activity of of R&D IL-6
• Measured in a cell proliferation assay using a factor dependent murine plasmacytoma cell line, T1165.85.2.1 (Nordan, R.P., Pumphrey, J.G., and B. Rudikoff. 1987. J. Immunol. 139:813).
• The ED50 for this effect is typically 0.2 - 0.8 ng/mL.
Formulation
• Lyophilized from a sterile-filtered solution in PBS.
Reconstitution of R&D IL-6
• It is recommended that sterile phosphate-buffered saline be added to the vial to prepare a working stock solution of no less than 100 μg/mL.
Storage of R&D IL-6
• Lyophilized samples are stable for up to twelve months from date of receipt at -20° C to -70° C.
• Upon reconstitution, this cytokine can be stored under sterile conditions at 2° - 8° C for one month or at -20° C to -70° C in a manual defrost freezer for three months without detectable loss of activity.
• Avoid repeated freeze-thaw cycles.
