Quantification of RNA in plant cells
I have been a loyal Custom Stellaris FISH probe customer for the last 4 years and have ordered over 10 sets. Every interaction I've had with the customer service team has been extremely helpful and their replies have always been prompt (given the time zone difference). I highly recommend the Stellaris probe designer. It is both straight forward and intuitive to use. Negative tests that I've carried out have always confirmed probe set specificity (although I recommend carrying out a BLAST check for each set before placing an order.)

The product seems to generally work as advertised, though it can be challenging to always understand how the Probe Designer generates custom probe sets, which can make troubleshooting and validation somewhat tricky at times.

Analyze gene expresssion pattern in single cell
The Stellaris RNA FISH experiment gives out stable and repeatable results. I think the drawbacks of those RNA probes is that the green fluorescence is always hard to detected and showed great background, which means the limitation of color combination. But if you only need to check two different probes, the probes are really good and easy to use.

Easy to order. The online application was user friendly. Only needed to input the sequence of the protein and the software took care of making the best probes for the sequence. Also, easy to use especially with the Stellaris FISH RNA kit.

RNA FISH to quantify/visualize lncRNA of interest
I did achieve high-quality results, with much less work than by traditional methods (ie northern, qPCR) plus with the bonus of spatial information. Other users should know that the difficulty of this method is directly proportional to the abundance of your target RNA. I did find the fluorophore nomenclature a bit different that I was used to, but all the resources to figure it out are on the website...

Stellaris FISH (fluorescence in situ hybridization) is a RNA visualization method that allows simultaneous detection, localization, and quantification of individual RNA molecules at the sub-cellular level in fixed samples using widefield fluorescence microscopy. A set of Stellaris FISH Probes comprises multiple oligonucleotides with different sequences each with a fluorescent label that collectively bind along the same target transcript to produce a punctate signal. The Stellaris FISH technology follows a simple protocol, does not use exotic reagents, is inexpensive and platform-independent, offers same-day results, and is versatile toward many sample types and applications. Scientists may even label Stellaris FISH probes with a number of various dyes to allow for multiplex detection of different RNA targets simultaneously. Finally, scientists can address the stochastic nature of gene expression and visualize RNA through direct detection without isolation, purification, and amplification.
