SURFE2R has proved itself as a valuable asset, providing robust and reproducible data

Average Rating 5.0

Application Area:

SSM-based Electrophysiology Screening Platform

SURFE²R 96SE is a unique platform and favors some exceptional and astonishing capabilities. Employing solid-supported membrane (SSM)-based electrophysiology, SURFE²R 96SE can resolve small currents of transporters and pumps in a high-throughput format that makes it ideal for screening assays. In our hands, SURFE2R 96SE has proved itself as a valuable asset, providing robust and reproducible data. Using SURFE2R 96SE, it is much more feasible to conduct concentration response screening in an industrial setting. Some targets such as organelle transporters that can be readily and in high-throughput format studied using SURFE²R 96SE, are inaccessible for other techniques.

Our "Gold Standard" as secondary assay system for electrogenic transporters.

Average Rating 5.0

Application Area:

Drug discovery, secondary transporter assays

The SUERFE2R is our "Gold Standard" as a secondary assay system for electrogenic transporters. Some experience is needed, as usual, for these kind of assay systems, but the instrument is easy to use and data evaluation is easy to conduct. The assay and technical support is superb.

The SURFE²R 96SE is designed for activity measurement of electrogenic membrane transporters and pumps, which generate currents too low to resolve by patch clamp or which are not expressed in membranes accessible for patch clamp (e.g. bacterial, organelles). The technology enables label- and radioactivity-free real time measurements with high time resolution.

The SURFE²R 96SE uses sensors in a standard 96-well plate format. Membranes containing the protein of interest are adsorbed onto these sensors. Any substrate can be added to activate the transporter. The resulting transport or binding currents will be detected and analyzed accordingly.

The whole process, from sample preparation and measurement to data analysis, is fully automated.

Features:

  • Recording of 96 wells in parallel using a 96-channel low-noise amplifier 10,000 data points per day
  • Automatization of experimental workflows including sample preparation
  • Automated data analysis and export, e.g. determination of EC50 8 positions for different solutions (1 well to 96 well)
  • More than 100 targets validated, 100 peer reviewed papers using SSM-based electrophysiology
  • Analysis of transporters, pumps and ligand gated channels
  • Measure even electroneutral exchangers and sugar binding
  • Use purified membranes and vesicles from cells or proteoliposomes
  • Label-free electrical measurements
  • Requires only 0.1 – 1 µg protein per sensor. This is sufficient for up to 100 experiments
  • Real-time data with high time resolution of 5 ms, not single point read-out
  • High signal amplification compared to patch-clamp
  • Fast binding kinetics can be resolved EC50, IC50, rate constants, comparison of transporter variants
