Application Area:

Pulldown of biotinylated proteins

I've been using these beads in my pulldown experiments over the years. They work quite well with high binding capacity. Biotin-streptavidin interaction is quite strong and it can resist harsh washing conditions. There is minimal sample loss, as long as you handle the beads carefully (not to touch the beads during washes etc). Usually, they settle down easily in low G spin but this may vary depending on the buffer you use. Magnetic ones could be another option if you have the proper rack- they are easier to settle down. However, since magnetic beads are much smaller, you can easily suck magnetic beads to the pipet tip so there might be more sample loss if you are not careful. Overall, it's a nice product.

Thermo Scientific Pierce Streptavidin Agarose is a standard-capacity beaded agarose resin of immobilized recombinant streptavidin protein for use in a variety of biotin-binding affinity purification methods.

Features of Streptavidin Agarose:

  • Streptavidin—purified recombinant streptavidin protein (53kDa, near-neutral pI); tetrameric with four biotin-binding site per molecule
  • Agarose resin—support is crosslinked 6% beaded agarose (CL-6B), the most popular resin for protein affinity purification methods
  • Inert and stable—streptavidin is immobilized by charge-free, leach-resistant covalent bonds, resulting in low nonspecific binding and enabling multiple uses without decline in yield
  • Standard capacity—this variety of beads has a normal load of immobilized streptavidin, providing a binding capacity of 1 to 3 mg of biotinylated BSA per mL of resin
