TrueBlue™ Peroxidase Substrate
KPL TrueBlue is up to 100 times more sensitive than DAB and it is free of known carcinogens. Strong blue reaction product provides excellent contrast with DAB and other substrates. KPL TrueBlue provides the most sensitive means available to detect peroxidase-labeled conjugates. Because of its high sensitivity, TrueBlue is recommended for detection of the least abundant marker. KPL TrueBlue is alcohol and xylene insoluble.

Immunocytochemistry
We have been using this for thousands of SARS-CoV-2 focus-forming assays over the past 9 months (virus titration and plasma neutralization assays). Loosely based on the method of Case et al. 2021. We use paraformaldehyde fixative, saponin/BSA perm-wash buffer, GenScript mAb A02058 anti-Spike primary, and Jackson anti-rabbit peroxidase secondary. This gives beautiful sharp dark blue/purple foci. There is very little background unless you didn't wash off the carboxymethylcellulose medium well before staining. In my previous job I have also used this substrate for dengue focus-forming assays.
