Western Blotting to examine the expression of proteins in cells, really good for PROTAC
Any scientist who runs Western Blots will know that the process is technically difficult, time-consuming, and requires 'good' antibodies that have very little cross-reactivity. The WES system from Protein Simple is an automated instrument that uses glass rods/tubes to form a column, then run the protein lysate to separate proteins based on molecular weight. In any one run either 12 or 24 lanes can be run, each lane contains your lysate plus fluorescence molecular weight markers that represent controls in each lane. It is also possible to multiplex the detection of more than one protein in each lane, allowing for a loading control in the same lane from the same sample. We have been using WES for over three years to detect proteins from recombinant cells, primary cells, etc. The system is very easy to use, allows a complete run to be achieved and analyzed in 3 hours and, although it is not as cheap as 'old fashioned' Western blotting, the data is highly quantitative and completely different from the traditional approach. Aurelia would highly recommend the WES.
