After years of working in neuropharmacology but far away from electrophysiology, we have had the opportunity to use the Patchliner. Our equipment is living a second life after 5 years in a box and after some software upgrades, it's alive and kicking! We only followed a day and a half of tailored formation at Nanion facilities in Munich and after 3 days we are having high-quality results, high productivity, and concentration-response curves after three mouse clicks! Just prepare the drug solutions and the suspension of cells and after 10 min you will get 6-8 concentration-response curves with their IC50 values automatically! Amazing! The patch liner is robust, reliable, and flexible. However, great equipment is nothing without support. We have received high-quality remote technical assistance that has solved all the small issues inherent to a new setup in a different lab and, most importantly, specific, high-quality scientific advice on which cells to use, what configuration to set up, experiment designs, and application notes, etc. A five stars team, because first of all, they are scientists and have a fair idea of what you need: electrophysiology without all the hassle.

Ion channels and human diseases
Our research focuses on understanding the pathology of ion channel disorders, using in vitro and in vivo electrophysiology to reveal opportunities for developing novel therapies. In particular, our research focuses on ion channel gene mutations that cause epilepsy. Patchliner has been an invaluable tool enabling us to examine ion channels both endogenously and exogenously expressed by a variety of different cell types, including mammalian cell lines as well as primary cells. The system allows us to examine the effects of different mutations and different compounds on the biophysical properties of a variety of ion channels, including sodium, potassium and calcium, to mention a few. Patchliner produces high fidelity, reproducible data which is very comparable to that produced using conventional techniques. It is easy to use and throughput is an attractive bonus. The systems temperature control is a really great feature for temperature sensitive channels. The after sales service from this company is second to none.

Drug discovery
Patchliner is an automated, single pipette, 8 parallel-channel system. It comes with many, easily modifiable, protocols for all ion channels groups. I have used Patchliners for primary, secondary and selectivity screens in voltage and ligand gated ion channel programs. The quality of the recordings is comparable to the manual patching, even for fast ligand gated channels such as alpha 7 nicotinic acetylcholine receptor. The technical support from Nanion is excellent. Love it.

Ion channel pharmacology
The Patchliner comes with robust and high quality Heka amplifiers. Therefore a previous knowledge of the Heka software is clearly helpful. Nevertheless, for the people that are not familiar with the Heka software, Nanion Patchliner created their own software, with a more friendly user interface, to control the Heka commands. Overall it is easy to use. The Patchliner offers the possibility to measure up to 8 cells simultaneously (some cells are "easier" than other though and the success rate can vary accordingly). The solutions are directly applied by the Patchliner "Syringe Pipettor". The speed of the solution application can be modified and the protocols are very flexible and easy to set up. Results are very reproducible. It allows the recording of all types of ion channels (including mechanosensitive channels!). In addition, the Patchliner support team is incredible to help you anytime you need in a very short notice!

The Patchliner is versatile yet robust, ideal for basic research of ion channel biophysics and mechanisms of action, sophisticated assays such as heat activation, right through to routine assays such as safety screening of hERG or other cardiac ion channels in accordance with the CiPA initiative.
The Patchliner is a medium throughput automated patch clamp system which comes as a 4 or 8 channel version. Experimental protocols are programmed using the PatchControlHT software which controls the Tecan robot for addition of solutions, cells and compounds, and controls the PatchMaster software for data acquisition. NPC-16 chips, the consumable for the Patchliner, can be loaded on the chip wagon and the instrument can run unattended for 48 recordings.
