IPTG (Isopropyl-beta-D-thiogalactoside) is an inducer of the lac operon and is frequently used in conjunction with X-GAL to determine the lac phenotype of recombinant plasmids or phage. IPTG is also used to induce the expression of cloned genes that are under control of the lac operon.
X-GAL (5-Bromo-4-Chloro-3 Indolyl-Beta-D-thiogalactopyranoside) is a chromogenic substrate for Beta-galactosidase that turns blue upon hydrolysis and is regularly used for the blue/white color selection assay. Bacteria with a lac+ phenotype grow as blue colonies on plates containing X-GAL.
