A Faster Solution with Increased Resolution for Determining Chromatographic Identity and Absence of OSCS in Heparin Sodium
17 Apr 2011The application note here demonstrates an improved method for resolving dermatan sulphate and oversulfated chondroitin sulfate in heparin with an IonPac AS11-HC (2 × 250 mm) column using a NaCl gradient followed by absorbance detection at 215 nm. Heparin is a complex highly sulphated glycosaminoglycan that has been used since the early 20th century as an anticoagulant for the treatment of thrombosis. The objectives here were to improve the resolution of DS from heparin, avoiding a perchlorate-based mobile phase, and reducing the analysis time. The method takes advantage of the high capacity and strong anion-exchange properties of the column to improve the resolution of the two contaminants in heparin.
