In this app note, Cytiva presents a workflow for membrane protein purification followed by detergent-free analysis. The traditional use of detergents for membrane solubilization is necessary for purification since membrane proteins are insoluble in aqueous solutions. The study's objective is to purify membrane proteins and subsequently reconstitute them into Salipro™ particles, where the proteins are surrounded by lipids instead of being in detergent micelles. The significance of this approach lies in its potential to enable Biacore™ analysis without the presence of detergents, allowing for the use of more native conditions in the analysis of membrane proteins.
