Biomarker Discovery and Identification by MALDI Molecular Tissue Imaging and Top-down Sequencing

This application note demonstrates the benefits of combining MALDI imaging and Top-down ETD/PTR techniques by using an amaZon ion trap as outlined in the Bruker PRIME (Proteomics through Integrated MALDI and ESI) concept. A novel, straightforward Top-down discovery and identification workflow for biomarker candidates has been developed. A peptide marker panel for HER2+ breast cancer was discovered and validated by MALDI tissue imaging. ETD/PTR on an amaZon ion trap was used for the subsequent Top-down protein identification.
