Determination of low-picomolar affinities of sgRNAs and crRNA/tracrRNAs for Cas9

Genome editing technologies based on CRISPR/Cas9 provide a plethora of applications in human therapeutics, food science and other fields. This application note demonstrates the versatility of MicroScale Thermophoresis to determine the binding affinities of various single-guide RNA, duplex of CRISPR RNA and trans-activating crRNA constructs with Cas9. The technology enables straightforward assay development, and optimal labeling strategy.
