Development of recombinant phage-like particles that encapsulates target RNA viral gene segments

This research poster presents the development of recombinant MS2 phage-like particles (PLPs) engineered to encapsulate target RNA viral gene segments as innovative positive controls for molecular diagnostics. Using a one-plasmid expression system, E. coli BL21 and T7 RNA polymerase drive maturation protein–coat protein fusion expression, enabling PLP production, purification via His-tag chromatography (AKTA Go), and validation through ddPCR, SDS-PAGE, Western blot, and TEM. The resulting PLPs demonstrate stability across multiple viral targets, including SARS-CoV-2 and FluA, offering scalable, biosafe controls for RNA-based diagnostics.
