In vitro protein synthesis of perdeuterated proteins for NMR studies

Perdeuteration of polypeptides is a useful technique for studying large polypeptides in solution, however this can be difficult to do in vivo. In this application note, Cambridge Isotope Laboratories summarizes work using cell-free synthesis methods employing both H₂O-based and ²H₂O-based E. coli cell-extracts for the non-uniform incorporation of ²H, ¹⁵N-labeled amino acids in vitro.
