Integrating magnetic bead-based antibody capture in high-cell density cultures

The production of monoclonal antibodies (mAbs) in high-density mammalian cell cultures is a cornerstone of modern biopharmaceutical manufacturing. Utilizing high-density perfusion, such as intensified fed-batch, cultures have gained prominence due to their ability to maintain consistent cell viability and productivity over extended periods. However challenges can be faced revolving high cell mass, especially at single harvest which is a major challenge for the downstream processing (DSP) in bioproduction.

Gain insights from a study that investigated the feasibility of using magnetic beads functionalized with Protein A to facilitate mAb capture from a high-density CHO (Chinese hamster ovary) cell culture in an intensified fed-batch bioreactor. The findings highlight magnetic separation as a scalable, efficient alternative for direct mAb capture from high-density cultures, offering significant advantages in purity, yield, and process efficiency.
