Modified sgRNA Design Improves Results of CRISPR Knockout Screens
9 Apr 2017Optimization of the sgRNA design used in CRISPR knockout screens has been the goal of several research groups in recent years. This application note aims to address points raised in recent studies, including whether modifications of the 80 bases downstream of the target region can increase the rate or knock-out efficiency of the active CRISPR system. Showing that it is possible to increase the quality of sgRNA libraries, with just a few changes to the 3' region.
