Stability optimization of engineered mAbs

In this application note, NanoTemper Technologies describes a study carried out by Merck to evaluate the stability of monoclonal antibodies (mAbs). The study evaluated two different families of mAbs with introduced mutations in their non-binding regions to determine how single and double mutations affected the structural core. The data revealed that even small changes to the protein sequence could have significant effects on the biophysical characteristics and stability of the mAbs. The note demonstrates the value of the simultaneous measurements of DLS, backreflection, and nanoDSF in characterizing the biophysical properties and stability of mAbs with mutations.
