An integrated multimodal analysis workflow to sort, sequence, and characterize antigen-specific B or T cells

B and T cells perform a vital function in the adaptive immune system. They recognize molecules the body has determined are foreign, called antigens, and subsequently trigger events that will clear those molecules and the pathogens they are derived from, such as viruses or bacteria. However, isolating antigen-specific B and T cells out of a complex sample with traditional techniques has proved challenging and time consuming for those in drug development. To solve this problem, 10x Genomics has developed a workflow to assemble antigens or peptides of interest with unique barcodes and fluorescent reporters, which can then be used for cell sorting.

This webinar will discuss the use of Barcode Enabled Antigen Mapping (BEAM), a multiplexed antigen screening workflow for empowering the rapid discovery of antigen-specific B-cell (BEAM-Ab) and T-cell (BEAM-T) clonotypes. The technology is built on the proven Chromium® Single Cell Immune Profiling solution. The unique workflow allows researchers to obtain comprehensive antigen-specific cellular profiles, including full-length paired V(D)J sequences, gene expression, and cell surface proteins, from the same single cell to accelerate therapeutic discovery.

Key learning objectives

  • Learn how to sort and characterize fluorescently labeled antigen-specific B or T cells with a customized gating strategy to enrich for binders and exclude non-binding populations
  • Understand how to generate tens to hundreds of high-quality antigen-specific hits from a single sample in one week
  • Learn how to capture extensive profiles of antigen-specific B and T cells, including full-length, paired V(D)J sequences, gene expression, and surface protein expression, at single-cell resolution
  • Discover rare and/or therapeutically relevant antigen-specific clonotypes to promote the next immunotherapeutic breakthrough

Who should attend?

Researchers who want to learn new approaches for successfully profiling antigen-specific T and B cells for high-throughput single-cell sequencing assays.

Certificate of attendance

All webinar participants can request a certificate of attendance, including a learning outcomes summary, for continuing education purposes.
