Size exclusion native mass spectrometry for the characterization of monoclonal antibodies
Size exclusion chromatography (SEC) is a well-established method for routine aggregate analysis of monoclonal antibodies (mAbs) and other related biotherapeutics. However, SEC can be combined with high-resolution mass spectrometry (HRMS) to obtain even more detailed characterization data for mAbs. So-called native SEC-MS techniques determine mAb higher order structure, relative abundance of glycoforms, and fragmentation.

Column selection, including pore volume and column dimensions, are also covered to ensure monomer and aggregate. Optimization of volatile mobile phase composition is shown for improvements in both MS sensitivity and chromatographic performance. Finally, methods for digestion of mAbs using site-specific proteases to obtain further insight on post-translational modifications will be described.
