Histone H3 is one of the core components of the nucleosome, the basic building block of chromatin. Histone H3 is subject to a variety of post-translational modifications (acetylation, methylation, phosphorylation) that affect packaging and accessibility of genomic DNA. Methylation of lysine 27 at histone H3 is carried out by the EZH2 methyltransferase, part of the Polycomb protein complex. The Polycomb complex silences gene expression by creating localized regions of facultative heterochromatin, and methylation of lysine 27 is part of this process.
