Description
Histone H3 is one of the core components of the nucleosome, the basic building block of chromatin. Methylation of histone H3 at lysine 4 is linked to transcriptional activation through the recruitment of a number of downstream effector proteins (e.g. CHD1, ING2, Isw1, NURF301, WDR5). Lysine 4 methylation is dependant upon the monoubiquitylation of the C-terminus of histone H2A.

Immunofluorescence
This antibody works very well by immunofluorescence in human cells at 1:200 to 1:400 dilution. Samples were first fixed and extracted for 10 minutes at room temperature in 4% formaldehyde and 0.1% Triton X100 in 1 x PBS before briefly blocking with 3% BSA in 1 x PBS-Tween for 30 minutes followed by 1 hour with the primary antibody (diluted in 1 x PBS-Tween, 1% BSA) and 1 hour with the secondary Alexa-488 conjugated secondary goat anti-rabbit (diluted in 1 x PBS-Tween, 1% BSA). Samples were post-fixed before application of anti-fade containing DAPI.

Immunofluorescence
This is a great antibody for detecting H3K4me2 in the nucleus of human tissue culture cells or on cytospun metaphase chromosomes. The background is low and the signal strength is high. We typically use this at a 1:200 dilution.
