Mili / PiwiL2 is a member of the PIWI/AGO subfamily of RNA-binding proteins. Mili / PiwiL2 contains a Piwi and an AGO domain and binds a class of RNA called piwi-interacting RNA (piRNA), which are 24 to 30 nucleotides in length and are derived primarily derived from transposons and other repeated sequence elements. Mili / PiwiL2 is required for methylation and inactivation of retrotransposons. Mice deficient in Mili / PiwiL2 are sterile, as it is also required for spermatogenesis. It also positively regulates translation, and is required for germ line stem cell division and differentiation.
