We use this antibody for two different techniques. Firstly, ex vivo labelling for flow cytometry. For this purpose, the Ly6G-PE antibody gives a good population of positive cells that can be clearly separated from the negative population. It is specific to the point that it allows detection of both the Ly6G-high and Ly6G-intemermediate cell populations. It is lowly expressed by some eosinophils which is worth noting, and for flow purposes for neutrophil analysis Siglec-F positive eosinophils needs to be gated out for analysis. This antibody goes well in a flow panel and identifies the neutrophil populations with a high specificity. For in vivo labelling and identification by intravital microscopy, this antibody works very well. It rapidly labels the circulating neutrophils when administered intravenously with minimal background and non-specific binding. It also copes well when cells are removed, fixed, and analysed by flow cytometry or imaging that is non-intravital.
