Description

KAP1 (KRAB Associated Protein 1, TIF1b, Transcriptional Intermediary factor 1b, Tripartite Motif-containing Protein 28, TRIM28, RING finger protein 96, RNF96) is an important co-repressor protein that plays a critical role in proliferation and differentiation of both normal and tumor cells. It functions as a co-repressor for KRAB domain-containing zinc finger proteins (KRAB-ZFPs) and mediates gene silencing by recruiting CHD3, a subunit of the Nucleosome Remodeling and Deacetylation (NuRD) complex. Part of its mechanism involves coordinating the increase in H3K9 methylation at promoters of silenced genes, potentially through direct recruitment of SETDB1, an H3 K9 methyltransferase involved in the formation of heterochromatin. KAP1 / TIF1b is also involved in maintaining pluripotency and is required for terminal differentiation of embryonic stem cells. KAP1 / TIF1b protein levels are increased in liver, gastric, lung, breast, and prostate cancer, and gastric cancer patients with high levels of KAP1 / TIF1b show a significantly lower survival rate. KAP1 / TIF1b contains two zinc fingers and a PHD finger.
