G protein-coupled receptors (GPCRs) play a crucial role in mediating cellular responses to external stimuli. BRET (bioluminescence resonance energy transfer) requires only the addition of a suitable substrate to generate donor light emission, as well as energy that can excite fluorescently-labelled ligands in a non-radiative manner. This application note demonstrates the first use of BRET for monitoring ligand binding to GPCRs in live cells enabled by NanoLuc® luciferase, using the CLARIOstar from BMG Labtech. Download method.

High-density peptide arrays have been measured with the Berthold Technologies bScreen featuring the Biametrics 1-lambda RIDe label-free technology. It is possible to screen up to 20,000 of peptide-protein interactions per cm2 within a few minutes and high affinity peptide binders against the pharmaceutically interesting target growth factor receptor-bound protein 2 (Grb2) can be identified and characterized. Antibody Drug Conjugates (ADCs) have been emerging as a new class of anti-cancer therapeutics, combining the targeted delivery capability of a monoclonal antibody (mAb) with the potency of conjugated cytotoxic agents. By releasing the cytotoxic drugs inside target tumor cells, ADCs minimize exposure of non-target cells to toxic drug payloads and thereby reduce potential side effects of the treatment. This application note demonstrates an integrated informatics workflow using UNIFI® Software, from Waters, that streamlines data acquisition, processing, and reporting for ADC analysis by LC-UV or LC-MS.
