The DRG:HYBRiD-XL 17-OH Progesterone is an enzyme immunoassay for the quantitative in vitro diagnostic measurement of 17-OH Progesterone (17-OHP) in serum andplasma.The DRG:HYBRiD-XL17-OH Progesterone is an enzyme immunoassay for the quantitative invitro diagnostic measurement of 17-OH Progesterone (17-OHP) in serum andplasma. Only for use with the DRG:HYBRiD-XL Analyzer.The steroid 17-Alpha-Hydroxyprogesterone(17-OHP) is produced by both the adrenal cortex and gonads. Even though 17-OHPhas relatively little progestational activity, it is of intense clinicalinterest because it is the immediate precursor to 11-desoxycortisol (Cpd-S).Because Cpd-S is produced by 21-hydroxylation of 17-OHP, measurement of 17-OHPis a useful indirect indicator of 21-hydroxy­lase activity. In congenital21-hydroxylase deficiency, the most common variety of congenital adrenalhyperplasia (CAH), 17-OHP is secreted in abundant excess. It is moderatelyelevated in the 11-Beta-hydroxylase deficiency as well.Measurement of 17-OHP is therefore valuable in the initial diagnosis of CAH.The DRG:HYBRiD-XL 17-OH Progesterone Kit is a solid phase enzyme-linkedimmunosorbent assay (ELISA) based on the principleof competitive binding. The antibody coated wells (ACW) of the reagent cartridges are coatedwith a polyclonal (rabbit) antibody directed towards a unique antigenic site ofthe 17-OHP molecule. Endogenous 17-OHPof a patient sample competes with a 17-OHP-horseradish peroxidase conjugate forbinding to the coated antibody. After incubation the unbound conjugate iswashed off. The amount of bound peroxidase conjugate is inversely proportional to theconcentration of 17-OHP in the sample. Having added the substrate solution, the intensityof colour developed is inversely proportional to the concentration of 17-OHP inthe patient sample.
