Proliferating cell nuclear antigen (PCNA) is a homotrimeric ring-shaped protein that encircles DNA and acts as a loading platform for multiple, transiently interacting factors participating in various DNA transactions. PCNA plays a key role in DNA replication, repair, cell cycle regulation, and post-replicative transactions like DNA methylation and chromatin remodeling. All these cellular processes are regulated by a complex network comprising cell cycle dependent changes in expression levels, dynamics, interactions, and localization of PCNA. ChromoTek’s Cell Cycle Toolbox offers the tools to investigate cellular processes in response to external stimuli, without interfering with regular cellular processes.
Cell Cycle Chromobody®
This Chromobody® comprises the antigen binding domain (VHH) derived from heavy chain antibodies of Alpacas genetically fused to TagGFP2 or TagRPF (Evrogen). Transfect your cell line of interest and:
• Trace the complete cell cycle in real time in live cells
• Monitor the distribution of an endogenous cell cycle marker protein - in comparison to fluorescent fusion proteins no overexpression artifact or cytotoxic effects
And simultaneously read out the following parameters:
• Cell size and morphology
• Nuclear morphology
• Progression of S phase (detailed detection of early-mid-late S phase)
• Mitosis
PCNA antibody (16D10)
Use the rat monoclonal PCNA antibody for the detection of denatured protein in applications like:
• Western blot
• IF
• IP
• Co-IP
