Painfully awful gels.

Gels stick to membrane after transfer. Bio-Rad know that they are faulty (people from a different institute got replacements). After several emails they sent replacements and they are still faulty. Can't get clear blots from these horrible gels. Switching to Invitrogen.

Analyse proteins in bacterial lysates

We have been ordering these precast gels for over 5 years in our lab as they save us a bit of time compared to making our own gels. Also, because the outer plates are disposable, we don't have to worry about risking breaking them, as with lab made gels which have fragile plates. There are cheaper precast gels out there but we have had the best results with these gels.

Western blots of various cancer cell line lysates

Really easy to use. These gels are nice because the lane for the ladder is small but the wells hold 45uL as opposed to 30uL so that if you need to load more lysate than normal or the lysate is dilute. We also use the Trans-Blot Turbo Transfer System. The transfer pads are a bit expensive but we are thinking about utilizing other transfer stacks to become more economical.

The Criterion™ TGX™ (Tris-Glycine eXtended) precast gels for PAGE are based on the long shelf life TGX formulation. The TGX gels retain Laemmli-like separation characteristics using the standard sample and Tris/Glycine running buffers. The proteins in the gel can be separated in as little as 20 min at 300 V while maintaining low temperatures.

Benefits Include:

  • Direct drop-in for Laemmli systems
  • Inexpensive Laemmli buffer system, low running costs
  • High-throughput system

TGX gels are manufactured without SDS. The gels can be used to run proteins under nondenaturing conditions for subsequent analysis of their native conformation and activity by omitting SDS from the running buffer. Native analysis does not provide accurate molecular weight information. SDS is added to the sample and running buffer to create standard SDS denaturing electrophoresis conditions.

Criterion gels are packaged and sold in individual units: 1–2 gels can be run in the Criterion cell and 1–12 gels can be run in the high-throughput Criterion™ Dodeca™ cell.
