The quality is of the highest standard. The support we get is unsurpassed, and fails to compare to any other company. We now see a welcomed trend towards GMP/CFR21 Part11 compliant, which will help us to incorporate the Wyatt instrumentation in our production processes. Especially for our outer membrane vesicles and conjugate vaccine development there is no other instrumentation that could help us to develop such vaccines to such high standard.

Characterization of antibodies, nanoparticles and extracellular vesicles

We've had the Eclipse AF4 flow field flow fractionation system in our research and development lab for over a year now. The system itself is very easy to use, especially if you have a basic understanding of HPLC and/or SEC. We have been able to obtain successful separation of samples containing both digested and undigested antibodies, and isolation of nanoparticles from multiple supernatant components. Wyatt customer service is great - every time we have run into an issue, we have gotten a response from support in less than 24 hours and they have always been able to solve our problem. The instrument is very expensive, but it is highly versatile and has become a key piece of instrumentation in our lab.

Field flow fractionation is a very difficult technique to master but the Wyatt Eclipse AF4 unit with included Scout predictive software, in addition to their 24/7 customer service, made method development and analyte characterization very achievable and enjoyable. Using one single instrument we have managed to separate and characterize polyelectrolytes with small radii from several nm up to hundreds of nm long rod like cellulose nanocrystals. The versatility and capability of this instrument is incredible and Wyatt's field reps must be credited for helping us realize the full potential of what AF4 can do.

Analyzing NP-particle interactions with organic colloids in environmental systems

With Wyatt Europe, I have a really good partner in after sales and application development/method development. The system is very flexible and with the service contract I always can count on immediate remote-help. Although, I am not very happy with the dictated combination of hardware and software developments meaning that I am somewhat dependend on expensive software-changes for optimal use of the hardware, and respective bug-fixes. As a university research institute we don't have the money to change working hardware, like in industrial settings. Wyatt US only provides online help if the instrumentation is not older than 10years, which means the possibility to satisfy customers like us is only possible by making a lot of effort. The free webinars offered to us during COVID19 were more than helpful, including the user seminars in the last weeks.

Nice and easy to use AF4 system. For an AF4 system it is amazingly robust and can be applied to a wide range of applications. The option to use a frit-inlet channel is useful to test for focus stress induced changes of your material.

The system works well for the purpose of study which is separation of exosome based on size. Although setup and optimization of instrument are quite complicated, the support staff are well-trained and professional. I usually get replies to emails email within 1-3 working days.

The instrument is designed well for ease of use in batch or being modular for online mode. However, the software needs to be well integrated so that it is a more cohesive software package where all of its capabilities are centralized into one package. As a result the software has a steep learning curve. Also, the AF4 channel design can definitly be improved with torqued latches so that assembly can be done more simplistically by flipping latches instead of manually by hand with a torque wrench.

Combined with Wyatt’s DAWN™ or miniDAWN™ multi-angle light scattering (MALS) detector and Optilab™ refractive index detector, the Eclipse offers a uniquely versatile FFF-MALS solution providing characterization of absolute molar mass, size, particle concentration, conformation and encapsulation.

Features:

  • Robust separation of macromolecules and nanoparticles from 1 nm - 1000 nm with excellent resolution. Typical analysis times are 20 - 60 minutes
