The multipurpose Gibco CTS Rotea Counterflow Centrifugation System offers flexibility in a cell therapy development and manufacturing system. This versatile system applies the proven counterflow centrifugation method to a broad range of cell processing applications, such as CAR-T therapy, stem cell therapy, and PBMC isolation. The CTS Rotea system consists of a compact multipurpose instrument, a sterile single-use kit, and intuitive, visual, customizable software with multiple software options to help enable 21 CFR Part 11 compliance. Together they constitute a closed system that offers exceptional cell recovery, flexible input and output volume capability, and high throughput. It’s ideal for multiple processes in your workflow, including cell separation, concentration, washing, and buffer exchange. Highly concentrated outputs (with volumes as low as 5 mL) contain very little buffer or carryover, optimal for autologous cell therapy applications. The compact footprint and closed-loop flow allow the system to seamlessly scale from process development through to commercial manufacturing.

The CTS Rotea Counterflow Centrifugation instrument, single-use kit, and CTS Rotea software work together seamlessly to provide a closed cell processing system. The CTS Rotea instrument is compact and versatile and integrates smoothly at multiple points in your workflow. The single-use kit includes all tubing and the rotating centrifuge chamber. Compatible with Luer connectors and sterile welding to the fluid containers, the gamma-irradiated kits help to ensure a closed system. Finally, the CTS Rotea software runs on an adjacent Dell™ system, validated by Thermo Fisher Scientific for use with the Rotea system, allowing you to view and modify protocols and visually monitor the process in real-time. The CTS Rotea system separates cells using the principle of counterflow centrifugation (CFC), a versatile method for separating cells based on their sedimentation characteristics (size and density). The system suspends cells in a fluidized bed by exerting a constant flow force against centrifugal forces. The fluid-suspended cells are gently concentrated without ever forming a pellet in the cone and washed efficiently at unmatched recovery rates. Using elutriation, dead cells can be removed to optimize viability. Cell types of different sizes and densities can be separated with the CTS Rotea system by tuning centrifugal speed and flow rate to create an imbalance of forces. The CTS Rotea CFC process is powerful yet extremely gentle on cells compared to other separation methods. The fluidized bed supports low-shear processing, enabling over 95% cell recovery while maintaining cell viability and achieving high throughput. Research shows that cell morphology and viability of cells are similar before and after CFC, confirming that the process minimally perturbs cells.
