The GS FLX Titanium Sequencing Kit XL+ combined with the GS FLX Titanium PicoTiterPlate Kit 70×75 provide reagents and other consumables required for determining the nucleotide sequence (in extra-long reads; up to 1 kb) of an immobilized and clonally amplified DNA library (the sample). The sample will have been originally prepared using the GS FLX+ System methods for the preparation of DNA libraries and amplified by the emPCR amplification process, using the appropriate GS FLX Titanium series kit(s). After amplification, the DNA-carrying beads are deposited into the wells of a PicoTiterPlate (PTP) device, such that each well contains no more than a single DNA bead. The loaded PTP device is then inserted into the GS FLX+ Instrument, and sequencing reagents are sequentially flowed over the plate. Information from all the wells in the PTP device is captured simultaneously by the camera, and processed in real time by the onboard computer. The raw data consists of a set of digital images from which the sequence of the DNA library fragments is determined, and then processed as appropriate for the sample type (e.g., assembled into a consensus). The whole sequencing process can be performed by a single individual in just a few hours, in a suitably equipped laboratory.
