The Instrument for Biomolecular Interaction Sensing by imaging Surface Plasmon Resonance (the IBIS-iSPR) is a high resolution SPR-imager that allows for simultaneous detection of hundreds of biomolecular interactions on a single sensor surface. One can see really in real-time ‘what’s going on’! Like a microscope one can view images of the entire sensor surface. Inspection of a microarray which is spotted on the sensor surface can be carried out instantly. A liquid handling system connected to a flow-cell is used to automate injections of many samples from 96 well microtiter plates.
Hundreds of Regions of Interest (> 500 RoI) can be freely defined (size and position) of which the user determines the association and dissociation rates of biomolecules on e.g. spotted surfaces of a microarray. Ligands can be loaded in different concentrations for single injection kinetic rate constant analysis. Because the IBIS-iSPR measures the real SPR angle shift from a RoI (and not only reflectivity) extremely high reproducible and accurate results can be obtained. The software includes a zoom-in function that facilitates RoI-definition on (patterned) sensors with small features. Contrasts can be maximized and even be virtually translated to high resolution color images.
