I had problems with a Cloning PCR of an GC-rich template, where I always got mismatches or small deletions in the GC-rich region with our conventional proof reading polymerase. With the KOD I got the correct clone in the first run already. Further I got a stronger PCR- band, respectively more PCR-product with virtually no background than with our conventional polymerase. KOD Hot Start combines the high fidelity, fast extension speed, and outstanding processivity of KOD with the high specificity of an antibody-mediated hot start. Non-specific amplification is reduced because mispriming events that can occur during setup and initial temperature increase are avoided. In addition, primer degradation during setup at room temperature due to exonuclease activity is effectively inhibited. KOD Hot Start Master Mix is a ready-to-use 2x mixture optimized for convenient high-fidelity PCR. KOD Hot Start DNA Polymerase generates blunt-ended PCR products.
