THE real time machine - forget the others

We also use it, alongside a BioRad CFX96 and an ABI 7500. Out of the three, I think the Roche has the best sensitivity, and it also allows you to buy a 384 well block that can be easily interchanged if you want to add high throughput capability. However, the software (or at least the version that we have) is not particularly quick, cannot be loaded on a personal computer, and doesn't have many options for ways to export the data. And when we were interested in expanding into using HRM, we had to cough up a lot of cash to get the upgraded software module which we haven't found to be particularly useful. In contrast, the BioRad CFX is our go-to machine for most 96 well applications. The machine is good, consistent, has never needed maintenance (even though it's often used 6x per day), and has faster ramp times. But the real boon is the software. The basic software is simple to use, can be loaded on everyone's computer for offline analysis, and offers even the difficult normalization options (like normalization to the geometric mean of several reference genes, or ways to normalize across several plates in a multi-plate experiment). It even offers plate wide melt calibration so it gives MUCH more consistent results for anything reliant on melt (like HRM). On balance, the Roche instrument doesn't seem anywhere near superior enough to the BioRad to justify the terrible software. I'd highly recommend taking a look at the BioRad CFX96. It's the one that gets the most use around here!

The Roche LightCycler 480 is a PCR instrument that uses either 96 or 384 well plates to carry out automatic measurements on gene expression (or microRNA expression). The instrument can be easily used. It uses melting temperatures to determine the quality and purity of the template. The machine can be serviced by Roche in Indianapolis, IN, and is reasonably priced. I am a fan of Roche and I recommend this product for accurate measurement of gene or micRNA expression.

It is very easy to perform optimization experiments and the software is easy to use. moreover, The LightCycler 480 PCR System utilizes a novel block technology to allow the rapid heating and cooling required to complete a 40-cycle, 384-well quantitative PCR run in less than 40 minutes and leads to extraordinary well-to-well temperature homogeneity and maximized inter-well, inter-cycle reproducibility. Finally, we suggest this system because the compatibility for automated sample-preparation, automated loading and connectivity to laboratory information management systems (LIMS) for integrated workflow monitoring offer uncompromising adaptability for a perfect fit.
