Purification of complex DNA extracts (e.g ancient DNA)
The OneStep™ PCR Inhibitor Removal Kit form Zymo is a great aid when working with complex and generally "dirty" DNA extracts. The usage is as simple an straight forward as it can be. Fast and accurate. The protocol and product description is great. I've recently started using it and I'm finding it extremely useful to clean up my extracts before library construction!

Remove inhibitors from peatland soil
It is very easy to use. The protocol explains for itself and the whole process takes less than 5 min. It removes the inhibitors effectively proved by DNase I digestion afterwards. The sales help a lot on building the RNA extraction pipeline with this kit and another kit also from Zymo, RNA clean and concentrator. This is the best inhibitor removal kit I have ever used.

Removing PCR inhibitors from DNA extraction
The PCR inhibitor removal kit is essential for some of the extractions we do on seaweeds. Our PCR products don’t work sometimes without this, so if you have samples with lots of secondary metabolites- like seaweeds, and the extraction kits used do not implement it already- you must get this.

Not worth the time or money.
Clean up RNA for qPCR
I tried using these PCR clean-up columns three times, with three samples each time. Each time, at least one of the columns failed during the first step where you run prep buffer through them. The material in the column would end up in the collection tube instead of forming the filter. I ended up using 4 or 5 columns for three samples each time. This step is also a 3-minute spin which adds up when two of them fail in a row and you are already pressed for time. The columns did relieve a bit of PCR inhibition, but were no more effective than simply diluting my samples (a much quicker and cheaper solution).

Remove inhibitors from DNA template for downstream PCR use
This product is super easy to use and the process is very quick. That's especially nice with a high volume of samples, which we have. We used this cleanup kit on DNA samples with poor 260/280 ratios, and it did help to improve that ratio.

Environmental DNA (eDNA)
Environmental DNA is full of inhibitors but working on a rare species means that nucleic acid retention is really important. These columns are effective in removing inhibitors while also retaining yield more effectively than silica clean up columns. The protocol is less involved, saving time, and uses less harsh chemicals, reducing exposure. I also really appreciate that Zymo offer sample kits for their products, this has really helped me in the important optimisation stages of my workflows.

The column matrices have been specifically designed for the efficient removal of polyphenolic compounds, humic/fulvic acids, tannins, melanin, etc. from the most impure DNA and RNA preparations. Sample clean-up is as simple as applying, spinning, and recovering a sample from the column.
