Separation of blood mononuclear cells
I've started using OptiPrep to separate monocytes from other lymphocytes in cord/peripheral blood. This product is great - it does take a little while to get the layering down with pipetting for the different densities, but the monocytes come out on top after centrifugation and are not activated prior to my usage (e.g., infections, etc). I would certainly recommend using this product, but as with all density gradient products, just be careful and slow when layering different densities on top of each other to make sure separations come out well after centrifugation.
