The Chorus has been a robust and versatile machine for our needs, regularly making a variety of linear and cyclic ~10 to 30mer peptides by Fmoc-based SPPS in an academic research setting. The synthesis procedures, reagents, temperatures, synthesis scale and resin usage are all highly customizable - giving great control over reactions, with single-shot minimal volume additions possible for expensive reagents. The ability to conduct several syntheses in parallel with automatic cleavage, all contained within the unit, really helps with throughput and safety to give us back time to do other things. The ongoing support from the sales and engineer team has been excellent, with any issues seen to swiftly and successfully, and GPT are receptive to feedback for continually improving the already solid software interface. We have enjoyed using it and getting great quality peptides out of it, would definitely recommend it.

We acquired the PurePep Chorus Automatic Peptide Synthesizer 6 months ago. Our laboratory is new to the peptide synthesis world and Gyros Protein Technologies has been with us since the beginning of this journey. We had a really good advice when choosing the equipment that best suited our current and future needs and the after-sales service has also been excellent. It must be said that, even though GPT is not present in Spain and the pandemic situation that we are experiencing since last year, we have not lacked the support of either the engineers or the experts at any time, and this is much appreciated. So far, the PurePep Chorus has been an excellent option for us because it is a very versatile device that will allow us to grow in the future. I would also like to emphasize that it is a very user-friendly instrument but without neglecting its robustness and reliability. We were looking for a peptide synthesizer that would allow us to perform automated parallel solid phase aromatic foldamer synthesis (SPFS). After a joint application-development with the team of Protein Technologies in Tucson, we acquired the PurePrep Chorus synthesizer about 6 months ago. Our coupling chemistry is more delicate than conventional peptide procedures and requires a pre-activation step to form the Fmoc-aromatic monomer (AM) acid chloride which is then coupled to the resin-bound aromatic amine. This pre-activation step necessitates anhydrous conditions due to the moisture-sensitivity of the Fmoc-AM-acid chloride. The Chorus fulfils these requirements. The acid chloride is formed in a pre-activation vessel (PV) prior to its transfer to the reaction vessel (RV) containing the resin. The synthesizer is now used routinely in our lab. We can build up to three foldamers either sequentially or in parallel depending on the protocols we wish to apply. The collect option makes it possible to recover our valuable monomers after coupling, which is really great. The Chorus is compatible with a large variety of solvents less common in peptide synthesis ranging from THF to chloroform, and it does not require complicated calibration steps. Due to the metered delivery of reagent solutions, the exact number of equivalents of coupling agents, additives and bases are delivered in the PV which makes our automated coupling chemistry reliable and repeatable. Heating by induction proves to be efficient. The targeted temperature is reached in a very short time (10-20 sec) and remains stable along the course of the coupling step. The new software is user-friendly and sequence programing is easy. We have already synthesized foldamers containing up to 30-aromatic units (equivalent to a 60-mer a-peptide in terms of backbone length) in 1.5 day. In summary, I highly recommend the Chorus synthesizer for SPFS and warmly acknowledge the Gyros-Protein technology’s team for their fruitful scientific exchanges.

We have been using the PurePrep Chorus peptide synthesizer from Gyros Protein Technologies for about 6 months now, synthesizing approximately 15 peptides per week. In our hands the system is an effective and efficient in terms of automated peptide synthesis. Furthermore, it is capable of synthesizing multiple (up to 6) peptides in parallel or sequentially when running different coupling chemistries. Compared to the PreludeX the improved software allows operating the instrument more intuitively and an upgraded database setup makes the system faster and improved the handling. Further, Gyros Protein Technologies has reduced on the consumption of solvents, reagents and amino acid solution resulting in reduced running costs compared to other synthesizers. Compared to the PreludeX, the PurePrep Chorus system is also less noisy while mixing. The instruments maintenance and after-sales-care provide by the skilled Gyros Protein Technologies engineers were/are excellent and we are very satisfied with the customer service. In the rare case of errors, Gyros Protein Technologies was very responsive and additional assistance was provided instantly. Overall, we can highly recommend the instruments provided by Gyros Protein Technologies, especially the latest PurePrep Chorus peptide synthesizer.
